    Assessment of acute-phase protein response associated with the different pathological forms of bovine paratuberculosis

    11 páginas, 1 tabla.In this study, the concentrations of two acute-phase proteins (APPs), haptoglobin (Hp) and serum amyloid A (SAA), were quantitatively assessed in serum samples from cattle naturally infected with paratuberculosis (PTB). APP profiles were compared across 190 animals classified according to the different pathological forms associated with infection: uninfected (n = 59), with focal lesions (n = 73), multifocal lesions (n = 19), and diffuse paucibacillary (n = 11) and diffuse multibacillary lesions (n = 28). Our results showed a significant increase in both APPs in infected animals compared to the control group, with differences depending on the type of lesion. Hp and SAA levels were increased significantly in all infected animals, except in cows with diffuse multibacillary lesions that showed similar values to non-infected animals. The expression pattern of both APPs was similar and negatively correlated with the antibody levels against PTB. These results indicate that the release of Hp and SAA is related to the presence of PTB lesions associated with a high cell-mediated immune response and a lower bacterial load, suggesting that the pro-inflammatory cytokines that are associated with these forms are the main stimulus for their synthesis. These molecules could show some potential to be used as putative biomarkers of PTB infection, particularly for the identification of subclinical animals showing pathological forms related to latency or resistance to the development of advanced lesions.This study was funded by grants RTI2018-099496-B-I00 from the Spanish Ministry of Science and Innovation and LE259/P18 from “Junta de Castilla y León”

    Analysis of local and peripheral immune response developed in sheep experimentally infected with Toxoplasma gondii at different times of gestation

(11-14 October 2017 - Madrid, Spain).Toxoplasmosis is one the main infectious causes of reproductive failure in sheep where the time of gestation when sheep are infected affects the clinical and lesional outcome of the disease. In order to investigate the variations of local and peripheral immune responses during gestation, pregnant sheep were infected at early, mid and late gestation and subsequently culled at 2, 3 and 4 weeks post infection. In those sheep inoculated during the second term, serological antibodies were detected earlier and the increase in serological γ-IFN was higher than in the other infected animals. Regarding the local immune response at the placenta, infiltration of inflammatory cells was mainly found in the maternal septa, although it also invaded foetal mesenchyme adjacent to the lesions. The increase in the number of T lymphocytes was observed only in ewes infected during the second and last terms of gestation while the increase of B cells occurred in sheep infected at the first and second terms. The expression of iba-1 antigen by macrophages was more frequent after infection during the first term whereas macrophages expressing lysozyme, CD163 or calprotectin were more frequent in infections at mid-gestation. A significant increase in the transcription of γ-IFN, when compared to control animals, occurred after infections in the first and second term, while TNF-α and IL-10 transcription increased only in the second and last term, respectively. There were no differences when comparing transcription of cytokines between animals infected at different terms of gestation. This study shows that the time of gestation when infection occurs has a clear influence over the pathogenesis of ovine toxoplasmosis as a greater inflammatory response was found after inoculating sheep at the second term of gestation.     Calicophoron daubneyi (Paramphistomidae) in slaughtered cattle in Castilla y León (Spain)

    The prevalence and aetiology of natural paramphistomosis was investigated in cattle slaughtered in the Castilla y León region (Spain) over a 3 year-period. The overall prevalence of positive animals was 6.20%. The parasite burden per animal ranged from 8 to 8005 (median = 144) and the ruminal atrium had the highest parasite burden whereas the ruminal dorsal sac the lowest. The prevalence and parasite burden increased with age while these parameters were lower in cattle under intensive management. Calicophoron daubneyi was the only Paramphistomidae species identified using morphoanatomical, histological and molecular methods in the studied animals.This work was supported by grant LE023A10-2 fromJunta de Castilla y León (JCyL).
